Predicting and evaluating drugs for combination therapyGuo and colleagues present a method known as CM-Drug for the identification of combination drugs that can boost the efficacy of immune checkpoint blockade therapy. They validate this method using melanoma and lung cancer models in mice and explore one hit from their screen in further depth, the thyrotropin-releasing hormone (TRH) analogue taltirelin.

Second-generation M1-polarized CAR macrophagesInduced pluripotent stem (iPS) cell-derived macrophages (iMACs) are being used to engineer CAR macrophages for immunotherapy. Zhang and colleagues design second-generation macrophage-specific CARs by integrating CD3ζ and TIR domains, resulting in M1-polarized CAR-iMACs with increased antitumor functions.
